Lowdrugloadingefficiencyisoneofthemainobstacleshinderingtheapplicationofcontactlenses(CLs)asthecarrierforextendedoculardrugdelivery.Hereinthisstudy,asimpleandeffectivedrugloadingmethodbasedonsaltinducedmodulationwasproposedanddemon-stratedwithmechanismelucidation.Firstofall,usingpoly(2-hydroxyethylmethacrylate)(p-HEMA)asthecontactlensmaterial,betaxololhydrochloride,DiclofenacSodiumandBe-taxololBaseasthemodeldrugswithdifferentsolubility,influenceofsaltconcentration,salttype(sodiumsaltsofsulfate,chloride,andsulfocyanate)anddrugpropertiesintheloadingsolutionondrugloadingefficiencywasinvestigated.Mechanismofenhanceddrugloadingincontactlenswasfurtherexploredviastudyingtheinfluenceofsaltontheab-sorptionisotherm,drugsolubilityandwatercontentofCLs.ApplicabilityofthismethodtootherCLsmaterialswasalsoinvestigated.ItwasdemonstratedthatadjustingtheionicstrengthofloadingsolutionsresultedinsignificantincreaseofdrugloadinginCLs.Typeandconcentrationofthesaltsandsolubilityofthedrugwerethemainfactorsinfluencingenhancementratioofdrugloading.Themechanismforimproveddrugloadingwasrelatedtothereduceddrugsolubilityinloadingsolutionsandthereducedboundwatercontentincontactlenses.ModulationofdrugloadingbyadjustingionicstrengthwasalsoapplicabletootherCLsandthelighttransmittancewasnotaffected.Thismethodwasmoresuitableforsalt-formdrugswithhighsolubility.Insummary,adjustingionicstrengthofloadingso-lutionisaneconomicalandeffectivewaytoimprovedrugloadinginCLs,andthissimplemethodmayalsofindapplicationinotherhydrogelbaseddrugdeliverysystems.
