联合用药对雷贝拉唑药代动力学影响研究

摘要 目的：研究抗生素如阿莫西林、克拉霉素及左氧氟沙星等对雷贝拉唑在大白兔体内药代动力学影响，为联合用药提供更为合理的依据。方法：6只大白兔先行十二指肠插管，插管处为幽门下10 cm处，导管经皮下隧道固定于背部皮下。给药后不同时间点取血，高效液相色谱法检测血药浓度，行药代动力学分析。每种给药方式结束后均予以1 w的清洗期。给药取血时间点为0 min、l min、3 min、6 min、10 min、15 min、30 min、45 min、l h、1.5 h，每点取血2 mL。结果：各组间药动学参数AUC(0-∞)(mg/L) h，Tl/2z(h)，Tmax(h)，CLz(L/h/kg)及Cmax(mg/L)经统计分析处理雷贝拉唑与阿莫西林合用及雷贝拉唑与阿莫西林、克拉霉素合用时药代动力学参数与雷贝拉唑单用时无明显差异性，雷贝拉唑与克拉霉素合用及雷贝拉唑与左氧氟沙星合用时AUC(0-∞)(mg/L)h虽较单用雷贝拉唑时有所提高，但无统计学差异，其他药动学参数亦无明显变化。结论：在雷贝拉唑与抗生素联合用于Hp根除治疗时抗生素对雷贝拉唑药代动力学无明显影响，表明抗生素对雷贝拉唑药效学可能无明显影响。

Study on Pharmacokinetics of Combined on Rabeprazole

Abstract Objective:To study the antibiotic such as amoxicillin, clarithromycin and levo oxacin and so on rabeprazole pharmacokinetics in rabbits effect for combination therapy to provide more reasonable basis. Methods:Six rabbits were first duodenal intubation under the Department for H. 10cm, catheter back through a subcutaneous tunnel fixed to the skin. Differ-ent time points after administration of blood, high performance liquid chromatography plasma concentration, pharmacokinetic analysis of line. After each administration method were to be 1 week washout period. Time point of blood administration 0min, lmin, 3min, 6min, 10min, 15min, 30min, 45min, lh, 1.5h, each point blood 2ml.
